If you plan to only do protein assay at 280 nm, you could select an inexpensive fixed-wavelength or a variable-wavelength instrument with the desired level of sensitivity. A few years ago, you would have selected a spectrophotometer with a sodium lamp and a 280 nm filter, but the lamp life for deuterium lamps used in variable-wavelength systems has increased dramatically. The capability to look at wavelengths ranging from 190 to 400 nm might be important for other applications. If you anticipate the need to look at colored samples, you might want to buy an instrument with an optional mercury lamp to extend your wavelength capability to 700 nm.

Often, I came back to see the sheet laminated in plastic and hung above the instrument for operator review and to aid in training other users. A very few instrument companies maintain application laboratories that will develop new application protocols for a customer, sometimes for free but usually for a nominal fee. These protocols can save the customer research time and the need to purchase complex, expensive equipment for methods development, not to mention hiring a trained, experienced development chemist.

Interconnecting cables and a software-controlled computer can unite dedicated and modular systems containing components designed for external control. Either system’s capability is dependent on the ability to upgrade the software as components change or are added. Dedicated systems usually link users to a single supplier that may offer state-of-the art advances. Modular systems usually offer more flexible or replaceable 20 ANALYTICAL INSTRUMENT SPECIFICATIONS generic software to fit new capabilities, such as adding more sensitive detectors or gradient capability to an HPLC system.
